There will be a repeat of the 2015 final while Brazil, United States and Uruguay have also found out their fate for the summer tournament
Holders Chile will come up against Lionel Messi and Argentina in the Copa America Centenario group stages later this year.
The two South American giants have been drawn alongside each other in Group D for the special 16-team summer tournament, which will be hosted in the United States.
Chile claimed the Copa America on home soil last summer by defeating Argentina on penalties, with Arsenal star Alexis Sanchez netted the crucial kick.
Meanwhile, Messi's Barcelona team-mate Neymar is expected to line-up for Brazil in a group which includes Ecuador, Haiti and Peru.
And Luis Suarez will lead his Uruguay team as they face off against Mexico, Jamaica and Venezuela.
Hosts United States have been handed a challenging group alongside Colombia, Costa Rica and Paraguay.
Manager Jurgen Klismann is optimistic about his side's chances of progression from what could be a tricky group.
"It's doable. We had a similar kind of scenario in Brazil [World Cup 2014], and we went through," Klinsmann said.
"First or second with that group is both tremendously fine."
The United States and Colombia will play the tournament's opening fixture.
Two teams will advance from each group into the quarter-finals, before the tournament continues in a standard knockout format.
Klinsmann continued: "We all want to see the best players from South America. We can't wait to have James Rodriguez (of Colombia) here, Messi and Neymar and all these wonderful players showcasing themselves in the United States.
